MYD88 Antibodies

MyD88 antibodies are highly specific reagents used to detect and study the expression, localization, and function of the MyD88 protein. These antibodies are essential tools for researchers investigating innate immunity, inflammation, and immune signaling pathways, as well as diseases involving abnormal MyD88 activity.

Applications of MyD88 Antibodies

  • Western Blotting (WB): MyD88 antibodies are widely used in Western blot assays to detect the expression levels of MyD88 in cell and tissue lysates. This application is particularly important in studying MyD88-mediated signaling in response to immune challenges, such as bacterial or viral infections.
  • Immunohistochemistry (IHC): In IHC, MyD88 antibodies enable the localization of MyD88 expression in tissue sections. This is useful for studying immune responses in various tissues, especially in the context of inflammation or autoimmune diseases, where MyD88-mediated signaling may be dysregulated.
  • Immunoprecipitation (IP): MyD88 antibodies are used in immunoprecipitation experiments to isolate MyD88 and its associated protein complexes from cellular extracts. This technique is valuable for studying the interactions between MyD88 and other signaling proteins, such as IRAK4 (IL-1 receptor-associated kinase 4), and for understanding how these interactions regulate immune responses.
  • Immunofluorescence (IF): MyD88 antibodies conjugated to fluorophores can be used in immunofluorescence assays to visualize MyD88 within cells. This is useful for studying the subcellular localization of MyD88, such as its recruitment to the plasma membrane following TLR activation, and for investigating its role in immune cell signaling.

Key Functions of MyD88 in Immune Signaling

  • Toll-like Receptor (TLR) Signaling: MyD88 is the primary adaptor protein for most TLRs, except TLR3. Upon ligand binding, MyD88 is recruited to the TLRs, leading to the activation of downstream kinases (such as IRAK4 and IRAK1), which subsequently activate the NF-κB pathway, resulting in the production of pro-inflammatory cytokines like IL-6 and TNF-α.
  • Interleukin-1 Receptor (IL-1R) Signaling: MyD88 also mediates signaling through the IL-1 receptor, triggering similar downstream signaling cascades that lead to inflammation. This pathway plays a crucial role in the body’s response to infection and tissue damage.
  • Adaptive Immune Responses: MyD88 plays a role in bridging innate and adaptive immunity by influencing the maturation and activation of dendritic cells, which are essential for antigen presentation and the activation of T cells.

MyD88 antibodies are essential tools for studying innate immunity, inflammation, and the signaling pathways that regulate immune responses. Through their various applications, MyD88 antibodies provide valuable insights into how the immune system responds to pathogens and how dysregulation of MyD88 contributes to disease.
