NPM Antibodies

NPM (Nucleophosmin), also known as B23, is a multifunctional nucleolar phosphoprotein involved in various cellular processes, including ribosome biogenesis, centrosome duplication, cell cycle regulation, and stress responses. NPM is highly abundant in the nucleolus, where it interacts with ribosomal proteins and ribosomal RNA (rRNA), playing a crucial role in the assembly of ribosomes.

Applications of NPM Antibodies
  • Western Blotting: NPM antibodies are commonly used to detect NPM protein in cell lysates or tissue extracts. This application is crucial for studying NPM expression levels under different conditions, such as cell stress or cancer progression, and for identifying alterations in NPM protein structure or processing (e.g., in the case of the NPM1 mutation in AML).
  • Immunohistochemistry (IHC): NPM antibodies are applied to tissue sections to localize NPM in the nucleolus and cytoplasm. This technique is particularly useful in cancer diagnosis, especially in detecting the cytoplasmic accumulation of NPM in AML patients carrying the NPM1 mutation.
  • Immunocytochemistry (ICC): In cultured cells, NPM antibodies are used to visualize the subcellular localization of NPM, which is primarily nucleolar but can translocate to the cytoplasm in response to stress or oncogenic transformation. ICC studies help elucidate how NPM contributes to nucleolar function and its dynamic relocalization under different cellular conditions.

  • Ribosome Biogenesis: NPM is a key regulator of ribosome assembly in the nucleolus. Researchers use NPM antibodies to study how NPM facilitates the interaction between ribosomal RNA and ribosomal proteins and how this process is affected by cellular stress or cancer.
  • Cell Cycle and Centrosome Duplication: NPM is involved in centrosome duplication and plays a critical role in ensuring proper cell division. NPM antibodies are employed to investigate how NPM regulates centrosome dynamics and how dysregulation of NPM contributes to genomic instability and tumorigenesis.
  • Cancer Diagnostics and Therapeutics: NPM antibodies are used in cancer diagnostics, particularly for identifying AML patients with NPM1 mutations. Additionally, researchers are exploring NPM as a therapeutic target, and NPM antibodies are being developed for potential use in targeted therapies that disrupt its oncogenic functions.
